In vitro cultivation of Besnoitia caprae on epithelial monkey, canine and baby hamster kidney cell lines.

ABSTRACT

 

Namavari M, Namazi F, Oryan A, Mansourian M, Rahimian A, Tahamtan Y In vitro cultivation of Besnoitia caprae on epithelial monkey canine and baby hamster kidney cell lines, Onl J Vet Res., 26 (1):86-92, 2022. Epithelial monkey (Vero), canine (MDCK) and baby hamster (BHK21) kidney cell lines were inoculated with tachyzoites of Besnoitia caprae from BALB-c mice. Bradyzoites were detached from tissue cysts of an infected goat for transition to tachyzoites in mice then transferred onto cell lines. Only BHK21 cells inoculated with tachyzoites from the peritoneal cavity of mice supported parasites with highest replication. Highest growth for tachyzoites occurred in BHK21 without any replication in MDCK or Vero cells. Mice and chicken embryonated eggs were inoculated with tachyzoites obtained from BHK21 cell culture. Mice given 1×105 tachyzoites died, whereas those given 5×104 from BHK21 survived 60 DPI with 2 mice exhibiting alopecia and skin lesions. Microscopy of skin revealed mild necrosis of epidermis and follicular adnexa with some mononuclear cell infiltration. Skin from mice and chicken embryonated eggs infected with tachyzoites of BC-Pars revealed a single PCR of 755 bp. We find that that B. caprae can be culture in vitro on BHK21 cells.
