Serological and molecular analysis of Brucella melitensis and Brucella abortus in a human cohort (Iran)

ABSTRACT

 

Kargar M, Dehghani F, Dousti A, Handali S., Serological and molecular analysis of Brucella melitensis and Brucella abortus in a human cohort (Iran), Onl J Vet Res., 18(9): 743-749,  2014.  We report the prevalence of Brucella amongst 1500 nomads from Iran using ELISA and PCR. Samples were screened by the Rose-Bengal test and tittered with 2ME and Wrights test. Antibody titers against Brucella IgG and IgM and Hemi-Nested PCR on Bru-F Bru-R primers and Bm-F and RR primers to B. abortus and B. melitensis  respectively, were determined. Ten percent or 156 samples were positive to Brucella spp., but no correlation was found between positives with month of sampling, region, sex, or  type of contiguity.  Ninety three percent (145 samples) were positive for IgG and 121 (77.56%) for IgM. PCR showed that of 49 (3.26%) positive samples 79.59% were B. melitensis and (6.12%) B. abortus.  There was no significant correlation between ELISA and PCR tests.  The high prevalence of Brucellosis among Nomads in Fars, Iran, was probably due to life style.
