Characterization of caprine (Capra hircus) SLC11A1 gene: innate resistance to paratuberculosis

Recent advances in immunology have permitted the identification of genes, amongst which SLC11A1 (solute carrier family 11 member A1), is associated with resistance or sensitivity to specific diseases. The aim of this study was to determine the sequence and structure of caprine (Capra hircus) SLC11A1 gene and the association of its polymorphisms with innate resistance to Mycobacterium avium subsp. paratuberculosis (MAP). One sample of blood, serum and faeces was collected from each of 50 goats over the age of 2, from a herd of 400 adult animals with a long record of paratuberculosis. This material was used for the sequence and structure analysis of the SLC11A1 gene and the detection of MAP DNA and antibody using Real Time PCR and ELISA. The goat SLC11A1 gene coding sequence consisted of 2004 bases and presented a 1648-bases Open Reading Frame encoding a 548 residue protein. The 3' untranslated region of the targeted gene revealed 2 microsatellites consisting of a variable number of guanine-thymine repeats, named regions A and B. Statistical analysis of the results indicated a significant level of association between the B8 allele of the polymorphic region B and the presence of MAP DNA in the faeces. The results presented here suggest association of specific polymorphisms of the SLC11A1 gene with innate resistance/sensitivity of goats to paratuberculosis is possible.
