Prevalence of Cryptosporidium spp in sheep in AL-Taji, Iraq

ABSTRACT

 

Mohammed T. S. Al-Zubaidi Prevalence of Cryptosporidium spp in Sheep in AL-Taji, Iraq, Onl J Vet Res., 21(5):251-258, 2017. Prevalence of  Cryptosporidium spp oocysts in naturally infected sheep in Iraq is reported. Fecal samples (90) from 1-3 y old sheep from Al- Taji, Iraq during February and May 2016 were examined by lugol's iodine, modified Ziehl-Neelsen stain and PCR. Prevalence of Cryptosporidium spp infection was ~49% (44/90) by PCR,  ~38% (34/90) by Ziehl-Neelsen and only ~28% (25/90) by lugol's iodine tests. Highest prevalence occurred in sheep ˂ 6 months old at ~70% (14/20) and lowest in 25-36 month old ones at ~35% (8/23). About 58% (25/43) of diarrheic sheep shed Cryptosporidium oocysts in feces. DNA extracted with 7 cycles of freezing-thawing in liquid nitrogen yielded more DNA than 3 three cycles by deep freeze. Results suggest that the SSU rRNA–based PCR detected Cryptosporidium oocysts in stool samples with high sensitivity.
