Determination of optimum dosage schedules of danofloxacin following two extravascular routes of administration in buffalo calves (Bubalus bubalis)

ABSTRACT

 

Sappal RK, Sidhu PK, Chaudhary RK, Sandhu HS., Determination of optimum dosage schedules of danofloxacin following two extravascular routes of administration in buffalo calves (Bubalus bubalis), Onl J Vet Res., 19(2): 90-101, 2015. We report rational dosage schedules for danofloxacin by linking in vitro PD data with in vivo PK profile of buffalo calves. The PK of danofloxacin was evaluated in eight healthy male buffalo calves following intramuscular (IM) and subcutaneous (SC) administration of danofloxacin at the dose rate of 1.25 mg/kg body weight. After IM dosing, mean values for PK parameters were: clearance 0.519 L/kg/h; terminal half-life 4.64 h and volume of distribution 3.425 L/kg. Mean values for terminal half-life and clearance were 4.03 h and 0.608 L/kg/h; respectively, after SC administration. The mean area under the plasma concentration versus time curves (AUC) were 2.455 and 2.154 µgh/ml, respectively, after IM and SC dosing and for bioavailability means were 100% (IM) and 94% (SC). Integration of in vivo PK data with in vitro minimum inhibitory concentration (MIC) against a pathogenic strain of P. multocida provided mean values of AUC24h/MIC ratios of 30h and 27.5h, respectively, after IM and SC dosing of danofloxacin. After IM and SC dosing, Cmax/MIC ratios were 3.70, and 3.62, respectively. Dosage schedules of danofloxacin for P. multocida were 5.20 mg/kg and 6.10 mg/kg per day for IM and SC routes of administration, respectively.
