Single dilution ELISA for estimation of antibodies against foot and mouth disease virus.

ABSTRACT

Solid phase (SP) and liquid phase (LP) single dilution ELISAs were developed for detection of antibodies against Foot and Mouth Disease Virus for serotypes O, Asia 1 and subtype A22 separately. Antibody titres of 80 Cattle serum samples collected before and 21 days after vaccination with polyvalent vaccine from an organized farm were determined by multidilution ELISA. The percent inhibitions were calculated at two fold serum dilutions starting from 1:10 to 1:80. The maximum coefficient of correlation (r) between percent inhibitions at different serum dilutions and respective antibody titres, was found at 1:40 dilution for all the FMDV serotypes in both SP ELISA and LP ELISA except for type Asia 1 LP ELISA, where the highest ‘r’ value was observed at 1:80 dilution.  Therefore, the percent inhibitions recorded at these serum dilutions were used further for calculating the parameters a and b of regression equation Y= a + bx. The regression equations for SP ELISA Y= 0.42+0.0238x, Y= 0.565+0.021x and Y= 0.678+0.0195x were derived for type O, subtype A22, type Asia 1 respectively, whereas for LP ELISA, regression equations Y= 1.1548+0.0104x, Y= 1.18+0.0081x and Y= 1.29+0.011x were derived for type O, subtype A22, type Asia 1 respectively. Serum antibody titres of 20 cattle serum samples estimated by multidilution and single dilution ELISAs, when compared statistically for SP and LP ELISAs revealed no significant difference between the antibody titres estimated by the two methods. These attributes make the single dilution ELISA, a test of choice for testing large number of samples economically at a faster speed.
