IL-1β and IL-8 mRNA proteins in equine bronchoalveolar fluids from obstruction due to straw inhalation.

ABSTRACT

We report IL-1β and IL-8 mRNA proteins by RT-PCR and western blot in broncho-alveolar lavage fluid (BALF) cytology in 8 horses during partial obstruction induced by straw inhalation. We used 4 susceptible horses fed on pasture as controls. Compared with controls, in those exposed to hay, within Compared with controls, in those exposed to hay, within the BALF neutrophils increased 61-83% and epithelial cells 63-71% whereas lymphocytes declined 21-45% (P  < 0.05). BALF neutrophils increased 61-83% and epithelial cells 63-71% whereas lymphocytes declined 21-45% (P  < 0.05). In BALF horses increases (P < 0.05) in IL-1β mRNA protein preceded increase of IL-8. Increased IL-1β protein, but not mRNA, correlated (P < 0.05) with BALF neutrophilia.  Increases in IL-8 mRNA and protein were transient, but protein peak at day 8 followed BALF neutrophilia peak. The increase of IL-1β, paralleled by neutrophil and epithelial cell accumulation in the BALF, is indicative of the active production of the cytokine by these cells. This reinforces the pivotal role of cells other than the macrophages in driving airway inflammation during RAO.
