 Effect of metformin and ripaglinide on histomorphology of polycystic ovaries in immature mice

Polycystic ovary syndrome (PCOS) is the most common cause of infertility due to anovulation in women. Insuline sensitive drugs are used to treat this condition. Effects of Metformine and Repaglinide on histomorphology of polycystic ovaries in mice is described. Polycystic ovaries were induced with 1mg testosterone enanthate daily in 5-7 week old groups of 8 mice each for 4 weeks. Mice were given 250 mg/kg metformin and 30 µgr repaglinide IP for 2 weeks. Eight PCO control mice were not treated. Ovarian weight/body weight ratio and diameter were measured. Microscopy of ovarian tissue with number of follicles were determined. Ovary/animal weight values in mice given metformin were lower (p <0.05) than controls whereas in those given Repaglinide there was no significant difference. Ovarian weight and diameter were greater in controls compared with those given metformin or repaglinide (p≤0.05) Average primordial, primary, cystic and atretic follicles in treatment groups were similar but significantly less compared with controls.  Preantral and antral follicles increased in mice given Metformin compared to other mice (p≤0.05). Results suggest that Metformin increased ovulation rates, growth of ovarian follicles and regeneration in polycystic ovary mice.
