receptor (MTNR1A) gene may affect seasonal reproduction in goats. We describe
single nucleotide polymorphism (SNPs) in goat MTNR1A gene exon 2 amplified by
PCR during breeding and non-breeding seasons. Jugular blood (5ml) was taken
into EDTA from 20 mixed breed non pregnant goats during breeding and non-breeding
seasons for DNA. Exon II of MTNR1A gene was amplified with a primer digested by
BsaI restriction enzyme to cleave 279 and 577 bp (C allele) and only one 856bp fragment (A allele) to
produce genotypes CC for 279 and 577bp, AA for 856bp and CA for 279, 577 and
856bp. Frequencies were 35% for genotype CC, 52.5% for CA and 12.5% for AA. By
Hardy-Weinberg allele frequency of C and A were 0.6125 and 0.3875, respectively.
Amplified goat melatonin gene receptor 1A (MTNR1A) had 856bp from 4 homozygous CC, heterozygous
CA genotype and homozygous AA genotypes.
We found 99% sequence similarity with Capra
hircus Gene Bank ID: AF419334.1 with 1349 hits and mutations T154C, C190T, G213A. G218A, G238A, G277A,G292C, G352A,
T421C, C424T, C577T, C589A, G763A, G797A, C817T with no
polymorphism in non-seasonal homozygote CC genotypes at locus 52 melatonin
receptor type 1A (MTNR1A) gene.
