Effect of penicillamine on acute phase protein, inflammation and tissue during acute nanoparticle silver toxicity in mice.

ABSTRACT

 

Namazi F, Fazeli M, Jalaee SR, Zolghadri Y, Nezhad SM, Nazifi S., Effect of penicillamine on acute phase protein, inflammation and tissue during acute nanoparticle silver toxicity in mice, Onl J Vet Res., 20(8):571-577, 2016. LD50 of intraperitoneal (IP) nanoparticled silver in 4 groups of 6 BALB/c mice was determined. Effect of penicillamine on nano particle toxicity in 6 groups of 6 female BALB/c mice given 50 or 200 mg/kg concurrently, or 4 hours after was assessed. Six mice were used as untreated controls. Haptoglobin (Hp), serum amyloid A (SAA), tumor necrosis factor alpha (TNF-α), Interferon gamma (IFN-γ) and interleukin 6 (IL-6) were assessed. Mice given nanosilver showed moderate degeneration of hepatocytes with mononuclear cell infiltration and swelling, with interstitial nephritis, peribronchiolitis, bronchiolar associated lymphatic tissue, interstitial pneumonia with macrophages and lymphocytes infiltration in lumen of alveoli. No lesions were observed in brain or heart. Severity of lesions appeared less in mice given penicillamine and no lesions were found in controls. The LD50 for nanosilver was 1500 PPM and there were significant increases in Hp, SAA, TNF-α, IFN-γ and IL-6 in all groups except those receiving 200 mg/kg penicillamine or controls.
