Effect of insulin-like growth factor (IGF-1) on proliferation of ovine spermatogonia stem cells.

Solgi M, Rahimi-Feyli P, Nikousefat Z, Moghaddam A., Effect of insulin-like growth factor (IGF-1) on proliferation of ovine spermatogonia stem cells, Onl J Vet Res., 20(5):327-334, 2016.   Undifferentiated spermatogonia stem cells (SSCs) self-renew and differentiate to produce transgenic animals and improve reproductive efficiency in ruminants.  We describe dose-effect of insulin-like growth factor-1 (IGF-1) on In vitro proliferation of ovine spermatogonia stem cells. Spermatogonia and sertoli cells were isolated from testes of 2-3 month old lambs by two-step enzymatic digestion and cultured for 10 days with IGF-1 10, 40 and 100ng/ml in media or without IGF-1 as control. Cell proliferation was evaluated at 4, 7 and 10 days. Type A spermatogonia were identified by immuno-histochemical staining against PGP9.5 antigen.  Spermatogonia stem cells and surface area in culture with 100ng/mg IGF-1 at days 4, 7, 10 were greater (P <0.05) compared with other treatments or controls. Treatment with 40ng/ml IGF-1 was different with other groups (P <0.05) and 10ng/ml had no effect. Results suggest that 100 ng/ml IGF-1 in culture medium may affect proliferation of ovine spermatogonia stem cells.
