Effects of selected antidepressants and antipsychotics on bone marrow cell

micronuclei and chromosome aberrations in male Swiss albino mice.

Amitriptyline (AM), imipramine (IM), sertraline (SE) and paroxetine (PA) are widely used for depression especially in patients with schizophrenia and bipolar disorder. Recent studies suggest that these drugs may induce genetic toxicity. We injected groups of 10 Swiss male mice each, daily for 7 days with combinations or single per kg 10mg AM, 40mg IM, 10mg SE, 10mg PA, 30mg clozapine (CL) and 10mg quetiapine (QE) intraperitoneally (IP). SE was also given with AM, PA with QE and SE with PA. Controls were given nothing or injected 0.25ml saline IP. Mice were sacrificed after 24h of last injection for bone marrow micronuclei and chromosome aberrations. For each treatment, 5 animals were used and 5000 cells examined. In controls we found 2.54-2.36% micronuclei whereas in those given AM, 2.82%, IM 2.76, SE 2.86, PA 2.92, CL2.68 and QE 2.72% (P < 0.05). Given together, SE with AM increased (P < 0.01) micronuclei to 3.38%, PA with QE 3.14% and PA with SE 3.12%. In mice given AM, chromosomal aberrations were found in 17.4% cells, SE 19%, IM 16%, PA 18.6%, CL 15.0% and QE 17.6% whereas in controls only 11.6%. When SE was given with AM, aberrations were 31.2%, PA with QE 27.6% and PA with SE 26.2% (P ≤ 0.05).
