Polymorphism in TNP-2 gene and its association with seminal parameters of cattle and buffalo bulls.

ABSTRACT

 

Panigrahi SK, Yadav BR, Polymorphism in TNP-2 gene and its association with seminal parameters of cattle and buffalo bulls, Onl J Vet Res, 13 (1):86-93, 2009.  In the process of sperm maturation, major restructuring of somatic chromatin wherein histones are  replaced by a group of arginine and lysine-rich transition proteins (TPs); which are in turn replaced by protamines, occurs. Transition nuclear protein-2 (TNP-2) gene was analyzed by PCR-SSCP to detect polymorphism in Sahiwal, Murrah and Karan Fries bulls. Sequence analysis of TNP-2 gene of Murrah buffalo bulls revealed two single nucleotide polymorphisms (SNPs) at 182 bp and 186 bp position of intronic region. However, the gene sequences of Sahiwal and Karan-Fries bulls revealed no variation. The effect of this polymorphism was explored on individual motility, mass activity and maturation of spermatozoa. Analysis of variance indicates two variants of Murrah buffalo have no significant effect on maturation, mass activity and individual motility of spermatozoa. However, breed had a significant effect on mass activity and individual motility, but not on sperm maturation.
