Molecular and pathological characterization of a highly virulent infectious bursal disease virus (vvIBDV).

ABSTRACT

 

Ranjbar VR,  Mohammad A, Dadras H Molecular and pathological characterization of a highly virulent Infectious bursal disease virus (vvIBDV), Onl J Vet Res., 17(7):370-382, 2013.  Infectious bursal disease virus (IBDV) causes an acute highly contagious disease in young chickens. Strains of recently isolated IBD viruses from sick and dead birds of IBD field outbreaks were characterized. The partial region of VP2 gene from IBDV field isolates were sequenced to assess genetic characteristics, and select viruses for further experimental study. The nucleotide sequence of the RT-PCR fragment was determined and submitted to GenBank (ShirazIBD isolate; Acc.no. JX983160). Then, 10 day old embryonated SPF eggs were inoculated with a field isolate virus. At 7 days post-inoculation (PI), embryos were examined for gross IBD lesions and RT- PCR test. Haemorrhages were seen on the CAM, skin and toe joint of the embryos. Spleens were enlarged, pale and had small necrotic foci. These lesions were severe in the early passages of the virus. Twenty one days old commercial chickens were inoculated with positive IBDV field bursal sample (groupA), CAM and embryo homogenate (groupB) and distillated water for control group (groupC). The birds were sacrificed three days after inoculation and bursa were collected for RT-PCR and histopathological examinations. Produced bands by RT- PCR methods in the infected tissue samples including homogenized bursal samples, CAM and chicken embryos were about 743bp. Histological evaluation of the bursa showed marked bursal lymphoid necrosis and depletion in both of the infected groups. According to these results, the isolated virus was diagnosed as vvIBDV.
