Effect of n-methyl-n-nitroso urea on vascular endothelial growth factor mRNA in spleen of rats

Vascular endothelial growth factor (VEGF) gene induces angiogenesis. VGEF expression and its receptors have been found in lymphoma cells. We report transcription of VEGF in spleen of female Sprague Dawley rats given N-methyl-N-nitroso-urea (MNU). Groups of nine 6 week old rats each were gavaged 60mg/kg (1) or injected intraperitoneal MNU (2) 4 times over 14 days. Controls consisted of six rats. Spleens were excised from euthanized at 24 weeks for RNA extraction and transcription of VEGF by qRT-PCR. RNA was converted to cDNA followed by amplification by RT-PCR. Amplification of VEGF transcripts were normalized with housekeeping genes glyceraldehydes-3-phosphate dehydrogenase (GAPDH) and beta-actin. Relative transcription of VEGF in spleen of rats injected MNU was 360.38±23.25, which was 316 times higher (P < 0.05) than controls (1.14±0.08), whereas in those gavaged MNU, transcription was 44.97±1.07 or 39 times higher compared with controls. Results suggested that VEGF was highly transcribed when MNU gavaged.
